PE 22-28 is a synthetic derivative of the naturally occurring Spadin peptide that binds to TREK-1. TREK-1 is found in areas of the brain that control mood, memory, and learning. PE-22-28 is being investigated for applications including antidepressant activity, education, stroke recovery, and neurodegenerative diseases such as Alzheimer's disease.

Interestingly, shortened analogs of Spadin actually show better TREK-1 inhibition than spadin itself. PE-22-28 is the representative peptide for this group of synthetic Spadin analogs. PE-22-28 has been shown to be more stable and have improved antidepressant activity and neurogenic properties over the naturally occurring Spadin.

In 2010, a peptide consisting of 17 amino acid residues was developed and synthesized, called Spadin, which is short for "Sortilin-derived Peptide with Antidepressant properties", i.e., a peptide with antidepressant properties obtained from a sortilin derivative. Spadin has the following amino acid sequence H-Tyr-Ala-Pro-Leu-Pro-Arg-Trp-Ser-Gly-Pro-Ile-Gly-Val-Ser-Trp-Gly-Leu-Arg-OH. Experiments showed that four-day treatment with the Spadin peptide not only produced a strong antidepressant effect, but also increased hippocampal phosphorylation of the CREB protein and neurogenesis, which is considered a key marker of antidepressant action after chronic treatment with selective serotonin reuptake inhibitors. These data indicate that Spadin is a potential new generation antidepressant with a rapid onset of action. Spadin can be considered as the first identified antidepressant peptide. Increasing requirements for the effectiveness and safety of pharmacotherapy determine the emergence of more modern drugs, including psychotropic drugs, including antidepressants. Improving therapeutic approaches to the treatment of depression is challenging. Current methods of using antidepressants are not suitable for many people, and when they are effective, they require several weeks of use before a therapeutic effect can be observed. To date, the main goals of the new drug development strategy are to reduce the time delay before the onset of antidepressant activity and to significantly reduce side effects.

The results of the studies demonstrated that PE 22-28 and its derivatives are promising molecules that can be an alternative to the Spadin peptide in the treatment of depression.

PE 22-28 has a high potential for use in the treatment of depression, regulation of neuro- and synaptogenesis, with increased efficacy and rapid action, manifesting itself after 4 days, in comparison with currently used antidepressants, whose action begins to appear only after 2-4 weeks applications.
