THYMOPENTIN (Thymus polypeptides) is a synthetic pentapeptide with the amino acid sequence Arg-Lys-Asp-Val-Tyr, which is the active site of the natural thymopoietin hormone with immunomodulatory properties. It is a polypeptide of the thymus (thymus gland). Thymopentin is produced mainly by epithelial cells of the thymus cortex and bone marrow; however, it is not known whether this peptide is secreted or retained in the cell.
Thymus polypeptides - thymosins - were discovered by A. L. Goldstein and A. White in 1965. In 1973, a standardized preparation suitable for clinical trials was isolated from calf thymus - thymosin fraction 5 (TF5, thymosin F5). TF5 consists of 40–50 peptide components with a molecular weight of up to 15,000 Da. A number of major peptides have been isolated from TF5 and their biological activity has been studied.
Studies in the 1980s showed that timopentin is a very well tolerated drug when used according to recommended dosing regimens. Some patients complained of drowsiness, but it is suggested that this symptom may be related to the known effect of thymopentine on neuromuscular junctions. It is also able to correct the imbalance of the immune system.
Studies show that in vivo injections of thymopentine do not cause a significant increase in the production of TNF-α and NO, and therefore do not have a toxic effect on the body.
It has also been shown that timopentin is well tolerated when taken simultaneously with various prescribed drugs, which indicates the absence of interaction of timopentin with other drugs.
