Natural endogenous micropeptide HUMANIN and its derivatives protect cells from damage in neurodegenerative diseases, including Alzheimer's disease. The peptide is also active in ischemic heart disease, atherosclerotic plaques, eye diseases and in improving bone growth.
Humanin is a naturally occurring micro-peptide found in mitochondria. It protects cells from apoptosis and may be useful in staving off heart, eye, brain, and muscle diseases. Humanin is one of a select, small class of compounds targeting mitochondrial function and thus improving cellular energy efficiency and homeostasis.
Experiments using cultured cells have demonstrated that Humanin has both neuroprotective and cytoprotective effects, and experiments in rodents have shown that it has a protective effect in models of Alzheimer's disease, Huntington's disease, and stroke.
Humanin was originally identified as an endogenous peptide that protects neuronal cells from apoptosis caused by various types of lesions associated with Alzheimer's disease. The researchers hypothesized that this peptide could protect energy deficient cells from mitochondrial disorders. An increase in humanin expression in skeletal muscle has been found in patients with mitochondrial encephalomyopathy with lactic acidosis and episodes of stroke.
In experiments the analogue of Humanin is able to normalize the concentration of glucose and reduce the symptoms of diabetes. Deterioration of insulin action is a metabolic hallmark of aging and is implicated in the development of age-related diseases, including type 2 diabetes mellitus and Alzheimer's disease. Studies have shown Humanin affects insulin sensitivity.
Experimentally, the effect of Humanin on the retinal epithelium was found, and it was found that Humanin nanoparticles are an effective tool for reducing the pathological factors characteristic of age-related macular degeneration.
Patented is a method for treating patients suffering from diseases characterized by tissue necrosis by administering to the patient a necrosis inhibitor and/or humanin at a dosage of 50–150 mg/kg body weight/day.
Data on the connection of Humanin with oncogenesis and cancer metastasis look contradictory. Obviously, further preclinical studies of the effects of the peptide in oncological diseases are required.
Humanin and its derivatives have a wide range of cytoprotective effects, which suggests the potential usefulness of the peptide in the development of drugs for the treatment of Alzheimer's disease and other neurodegenerative diseases, diabetes, diseases of the heart, eyes, and others.
